厚朴酚调控人小细胞肺癌H446细胞凋亡的机制研究 被引量:4

Study on the Mechanism of H446 Cells Apoptosis of Magnolol-Induced Human Small Cell Lung Cancer

摘要 目的:探讨厚朴酚对人小细胞肺癌H446细胞增殖和凋亡的影响。方法:体外培养人小细胞肺癌H446细胞,经不同浓度和不同时间的厚朴酚作用后,应用MTT法检测厚朴酚对H446细胞增殖的影响;采用流式细胞仪罗丹明123染色法检测线粒体膜电位的改变;蛋白印迹方法检测凋亡相关蛋白的表达。结果:厚朴酚可呈时间剂量依赖性抑制H446细胞增殖,诱导线粒体膜电位的下降,免疫印迹结果显示Bcl-2的表达降低,Bax、Cyto.c和活化pro-caspase 9的表达增加。结论:厚朴酚可显著抑制H446细胞增殖,介导线粒体途径的细胞凋亡。 Objective:To study effects of magnolol on cell proliferation and apoptosis in human small cell lung cancer H446 cells in vitro.Methods:H446 cel1s were cultured with magnolol in different concentrations and for different periods in vitro.Cell growth inhibition was measured by MTT assay.The mitochondrial membrane potential was detected by flowcytometric analysis of Rhodamin 123 staining.Protein expression was detected by Westem blot assay.Results:Magnolol induced cell growth inhibition in a time-and dose-dependent manner.Compared with control group,magnolol-induced mitochondrial membrane potential collapse decreased the protein expression of Bc1-2 and increased the protein expressions of Bax,Cyto.c and pro-caspase 9.Conclusion:Magnolol significantly inhibited the proliferation of H446 cells and induced mitochondrial apoptotic pathway.
