摘要
用流式细胞术测定163例人鼻咽癌前与癌细胞DNA含量,用DNA指数(DNA index)均值表示相对含量。以正常人淋巴细胞及人胎儿鼻咽上皮细胞DI为正常对照(DI=1.000);炎症、单纯增生及轻度异型增生细胞均正常(DI=1.023);中、重度异型增生DI值分别为1.181和1.27;癌细胞DI均值高达1.332,其中鳞癌细胞分化越差,DI值越高,异倍体百分比越高,高分化鳞癌DI均值1.325,异倍体率61.29%;低分比鳞癌DI均值为1.460,异倍体率90%。未分化癌DI均值为1.426,异倍体率100%。说明细胞恶变过程中,细胞恶性程度与DNA量呈正相关。并比较了癌组织生长方式与DNA量的关系。
The morphological criteria of precancerous lesions used in the past does not answer the question whether the potency of neoplastic transformation exists. We used flow cytometry (FCM) technique to quantitate the DNA contents and the ploidy of precancerous cells to investigate whether it could be used in the diagnosis of the malignant potency.
One hundred sixty three samples were analysed. These included 2 samples of fetal normal nasopharyngeal epithelixm, 3 samples of normal human lymphocyte, 4 samples of simple hyperplasia, 28 samples of dysplasia and 126 samples of cancer of nasopharynx. The results showed that the DNA Index (DI) of the fetal epithelium and lymphocyte was 1.00±0.10 (Mean+RD) which was used as nornal value; the DI of the 4 samples of simple hyperplasia lesions was 1.023±0.19 and no aneuploidy was observed; the DI of 28 samples of dysplasia lesions was 1.118±0.137 arid aneuploidy was identified in 42.86% of the cases; the DI of the 126 samples of cancer was 1.330±0.19 and the aneuploidy rate was 77.78%. The DI value and the percantage of aneuploidy correlated to the differentiation of the cells, the poorer the differentiation of the cells, the higher the DI and percentage of aneuploipy.
