摘要
目的检测食管鳞癌中微小RNA miR-200c的表达,探讨其与肿瘤发展和分期的关系。方法应用实时荧光定量PCR方法检测76例各类分期分型食管鳞癌和配对癌旁组织中miR-200c的表达,分析其表达与肿瘤临床分期和病理分型的关系。结果食管鳞癌组织与癌旁组织比较,miR-200c的表达显著减少(P<0.01);miR-200c的表达在中晚期(III、IV期)食管鳞癌组织较早中期(Ⅰ、Ⅱ)食管鳞癌组织降低更为显著(P<0.01)。结论 miR-200c在食管鳞癌发生发展过程中表达有明显降低,其表达水平可作为潜在的食管鳞癌临床分期分级和预后的指标。
【Objective】 To examine the miR-200c expression in esophageal squamous cell cancer(ESCC) and correlate it with patient clinical status.【Methods】 Expression levels of miR-200c were quantitated with stem-loop quantitative reverse transcription-PCR in 76 ESCC patients and paired normal controls.The obtained results were correlated to the pathoclinical status and metastasis of the patients.【Results】 ①Expression levels of miR-200c were over-expressed in ESCC tissues compared with normal controls(P 〈0.001).② Statistics analysis showed that miR-200c was more amplified in advanced and low differentiated ESCCs(P 〈0.01).【Conclusion】 Amplification of miR-200c is frequent in ESCCs and is correlated with tumor progression.The expression level of miR-200c may be used as a potential biomarker of ESCC progression.
