高效液相色谱荧光法测定人血浆中顺式阿曲库铵浓度 被引量:1

摘要 目的建立测定人血浆中顺式阿曲库铵质量浓度的高效液相色谱荧光法。方法采用乙腈沉淀蛋白进行血样处理,色谱柱为Dia-monsil C18柱(150 mm×4.6 mm,5μm),流动相为0.04 mol/L磷酸二氢钾(pH 3.0)-乙腈(60∶40),流速为0.5 mL/min,荧光激发波长为230 nm,发射波长为324 nm。结果顺式阿曲库铵进样质量浓度在10.0~2 500 ng/mL范围内与峰面积线性关系良好(r=0.999 0,n=7),检测限和定量限分别为2.5 ng/mL和10 ng/mL,日内RSD小于3.6%,日间RSD小于4.0%,提取回收率为88.91%~91.09%,方法回收率为97.95%~98.70%。结论该方法简便、灵敏、准确、重现性好,适用于人体内顺式阿曲库铵药物代射动力学的研究。 Objective To establish a HPLC fluorescence method for the determination of cisatracurium in human plasma.Methods The blood sample was treated for the protein precipitation by acetonitrile.The separation was carried out on the Diamonsil C18 column(150 mm×4.6 mm,5 μm) with 0.04 mol/L potassium dihydrogen phosphate(pH3.0)-acetonitrile(60:40) as the mobile phase at the flow rate of 0.5 mL/min,and the excitation and emission wavelengths were 230 nm and 324 nm,respectively.Results The calibration curves were linear(r=0.999 0,n=7) in the range of 10.0-2 500 ng/mL for plasma cisatracurium.The limits of detection and quantification were 2.5 ng/mL and 10 ng/mL,respectively.The intra-day and inter-day precision were less than 3.6%and 4.0%,respectively.The mean extraction recovery rate was 91.1%,and the mean method recovery rate was 98.5%.Conclusion This method is simple,sensitive,accurate and better reproducible,which can be applied to the clinical pharmacokinetic investigation of cisatracurium.
