摘要
【目的】了解蔗糖磷酸合成酶基因在甘蔗中表达特性及其对甘蔗糖分积累影响的作用机理。【方法】以甘蔗品种桂糖28号(GT28)、拔地拉、新台糖20号(ROC20)、新台糖22号(ROC22)为供试材料,选取0叶、+1叶、幼嫩叶鞘、幼茎,通过半定量RT-PCR法对甘蔗SPSIII基因的表达进行分析。【结果】SPSIII在4个甘蔗基因型中的0叶、+1叶、幼嫩叶鞘和幼茎中均有表达,在GT284个不同部位的表达量较低,在拔地拉4个不同部位表达较均匀,在ROC204个部位中以嫩叶鞘的表达较高,在ROC22中以幼茎的表达较高。【结论】SPSIII在4个甘蔗基因型中的0叶和+1叶、幼嫩叶鞘和幼茎中均有表达,但SPSIII表达因甘蔗基因型不同而有所差异。
[Objective]The present studies were conducted in order to understand expression of sucrose phosphate synthase genes Ⅲ (SPSⅢ) in sugarcane and mechanism of its action during sucrose accumulation. [Method]Four sugarcane varieties, viz., GT28, Badila, ROC20 and ROC22 were used to sample leaver+l and 0, young sheath and young stalk for assaying expression of SPSⅢ using semi-quantitative RT-PCR. [Result ]The results obtained are as follows: SPSⅢ was expressed in all four sampled parts, i.e., leaf+l and 0, young sheath and young stalk in all sugarcane genotypes, but the expression levels varied in different genotypes. The expression of SPSⅢ was vetT low in all four parts of GT28, while it was higher in all parts of Badila, and showed highest in young sheath of ROC20 and young stalk of ROC22. [Conclusion ]It is concluded that SPSⅢ was expressed in leaves, young sheath and young stalk of all sugarcane varieties, however, its expression varied in different parts of different sugarcane genotypes.
