摘要
根据GenBank中已登录的多种植物的蔗糖磷酸合成酶基因的保守序列设计引物,采用RT-PCR方法,从枇杷果实中扩增出该基因的cDNA片段并测序,分别为386 bp和383 bp。序列分析表明,枇杷果实蔗糖磷酸合成酶基因与桃果实蔗糖磷酸合成酶基因相似性达91%。半定量RT-PCR分析表明,该基因在枇杷果实生长发育过程中的表达量逐渐增加。所得基因片段已在GenBank中注册,386 bp和383 bp登记号分别为HM122759和HM146926。
The 386 bp and 383 bp cDNA encoding sucrose phosphate synthase fragment were cloned from the fruit of loquat by the method of RT-PCR on the basis of the homologous genes of many plants in GenBank.Sequencing analysis showed that there was 91% identity in the nucleotide sequence of sucrose phosphate synthase gene between loquat fruit and peach fruit.Semi-quantitative RT-PCR analysis showed that the expression quantity of sucrose phosphate synthase was gradually increased during the growth and development of loquat fruit.The gene fragments have been registered in GenBank,the access number of 386 bp and 383 bp cDNA fragments is HM122759 and HM146926 respectively.
