摘要
鹿茸是唯一可以重复再生的哺乳动物器官。利用不同的提取方法制备梅花鹿鹿茸总蛋白样品,分别进行双向电泳,从得到的蛋白点的数量和点的圆滑程度、分辨率等确定适合的提取方法。分别用全蛋白提取试剂盒、自制裂解液、丙酮沉淀制备蛋白样品,并对得到的3种2-DE图谱进行比较分析。结果显示,试剂盒提取法和自制裂解液法得到的蛋白点比较多、圆滑,图谱清晰;丙酮沉淀法得到的蛋白点不如前者多,图谱有横竖纹。
Deer antlers are the only mammalian organs capable of repeated regeneration. To prepare total protein samples of sika deer antler for 2 DE by different extraction methods, and then find a suitable one from the spots number and resolution condition in the map. Total protein extraction kit, manual lysate and acetone precipitation were used and three kinds of 2-DE maps were analyzed. The results showed that spots number and resolution of kit and manual lysate were better than that of ac- etone precipitation, and stripes in the map of acetone precipitation were visible.
