摘要
解剖梅花鹿鹿茸,取得间充质,经过胰酶消化,体外培养鹿茸间充质细胞。利用TRIZOL提取鹿茸间充质细胞总RNA,利用BD SMART技术构建鹿茸间充质细胞全长cDNA文库,测定该文库初级文库滴度为3.0×105 pfu/mL,扩增文库滴度为6.0×108 pfu/mL。插入片段长度大于500 bp。
This experiment obtained the mesenchyme from the antler by the histology dissection.Then we digested the material with the trypsin and cultured the antler mesenchymal cells.Subsequently,total RNA was isolated form antler mesenchymal cells with TRIZOL,and a full-length cDNA library of antler mesenchymal cells was constructed.The titer of the primary library was up to 3.0×105 pfu / mL,after amplified,the titer was 6.0×108 pfu / mL.Insert fragments＇ length were more than 500 bp.
