摘要
目的 观察联合使用七氟醚和异丙酚对背根神经节细胞γ 氨基丁酸 (GABA)调控的氯电流影响。方法 采用酶解和研磨技术游离SD幼鼠背根神经节细胞 ,应用膜片钳全细胞记录技术记录氯电流 ,GABA采用压力喷射装置给予 ,溶解于细胞外液中的异丙酚、七氟醚则采用“Y”型管给药。结果 3× 10 -4 ,7× 10 -4 ,1 5× 10 -3 ,2 2× 10 -3 及 3× 10 -3 mol/L七氟醚分别增强氯电流至基础值的 149%± 2 5 % ,2 0 3%± 2 7% ,32 7%± 78% ,333%± 10 9%及 2 43%± 71%。 1× 10 -7,3× 10 -7,1×10 -6,3× 10 -6及 1× 10 -5mol/L异丙酚亦分别增强氯电流至基础值的 10 8%± 11% ,131%± 18% ,191%± 5 1% ,2 72 %± 48%及 789%± 86 %。二者联合使用后引发的氯电流高度与七氟醚、异丙酚分别产生的氯电流高度的理论之和的差异无显著性 (P >0 0 5 )。
Objective To investigate effect of the combination of sevoflurane and propofol on chloride current induced with γ aminobutyric acid(GABA) in the rat dorsal root ganglia neurons Methods The cultured rat dorsal root ganglionic neurons were dissociated by collagenase and trypsin, and trituration GABA 3 μmol/L was administered by pressure ejection The whole cell patch clamp technique was used to record the chloride current Propofol and sevoflurane dissolved in external solution and given through a rapid perfusion system named“Y tube"method Results With the peak current amplitude induced with GABA 3 μmol/L being regarded as 100%,the peak amplitude of chloride current increased to 149%±25%,203%±27%,327%±78%,333%±109% and 243%±71% following the administrations of sevoflurane at 3×10 -4 ,7×10 -4 ,1 5×10 -3 ,2 2×10 -3 and 3×10 -3 mol/L respectively, and rose to 108%±11%,131%±18%,191%±51%,272%±48% and 789%±86% in the presence of 1×10 -7 ,3×10 -7 ,1×10 -6 ,3×10 -6 and 1×10 -5 mol/L propofol respectively Following the combinations of sevoflurane (7×10 -4 mol/L) and propofol (3×10 -7 , 1×10 -6 or 3×10 -6 ), the induced peak amplitudes of chloride currents were not different from the theoretical additive values between sevoflurane and propofol (P>0 05) Conclusions Propofol combined with sevoflurane additively potentiates GABA gated chloride current in rat dorsal root ganglia
