摘要
【目的】建立同时测定银实润肠颗粒中绿原酸和柚皮苷含量的方法。【方法】采用高效液相色谱(HPLC)法,以Kromasil C18(4.6 mm×250 mm,5μm)色谱柱为固定相,以甲醇—体积分数0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL/min,柱温25℃,进样量10μL,检测波长为327 nm(绿原酸)、283 nm(柚皮苷)。【结果】绿原酸、柚皮苷的保留时间分别为9.53、30.12 min,两者浓度分别在9.7~77.6μg/mL、100.6~804.8μg/mL范围内线性关系良好,相关系数分别为0.999 8、0.999 9,平均加样回收率分别为99.01%、98.47%,sR分别为2.80%,2.42%。【结论】本研究方法操作简便,结果准确、可靠,重复性好,可用于银实润肠颗粒的质量控制。
Objective To establish double wavelength HPLC for simultaneously determination of the content of chlorogenic acid and naringin in Yinshi Runchang Granules. Methods HPLC was performed on a Kromasil C18column (4. 6mm ×250 mm, 5μm) at 25℃. The mobile phase consisted of methanol -0. 1% phosphoric acidsolution, and gradient elution was used. The flow rate was set at 1.0 mL · min^-1 and the ultraviolet detector was operated at 327 nm for chlorogenic acid and 283 nm for naringin, respectively. Results The retention time of chlorogenic acid was 9.53 min and that of naringin was 30. 12 min. The linear ranges of chlorogenic acid and naringin were 9.7 -77.6μg/mL and 100.6 -804. 8μg/mL, the average of sample recoveries were 99.01% and 98.47% , and the RSD (N =5) were 2. 80% and 2.42% , respectively. Conclusion The method shows good separation and the results are accurate and repeatable. It can be used for the quality control of Yinshi Runchang Granules.
