RP-HPLC法同时测定小建中颗粒中环磷酸腺苷和甘草酸的含量 被引量:1

摘要 目的:建立高效液相色谱法同时测定小建中颗粒中环磷酸腺苷和甘草酸含量。方法:采用Agilent TC-C18(4.6 mm×150 mm,5μm)色谱柱,流动相为甲醇(A)-0.01 mol.L-1磷酸二氢钾溶液(B),梯度洗脱(0~12 min,90%B;12~30 min,30%B),流速:1.0 mL.min-1,检测波长为250 nm,柱温为30℃。结果:环磷酸腺苷和甘草酸在12.5~200μg.ml-1质量浓度范围内与峰面积呈良好的线性关系,环磷酸腺苷和甘草酸的线性方程分别为:Y=23190X-4.6083(R2=0.9998),Y=11364X+80.342(R2=0.9950),环磷酸腺苷和甘草酸的回收率分别为95.2%和97.5%;RSD分别为2.8%和1.8%。结论:本方法操作简便,经方法验证可用于中成药小建中颗粒中环磷酸腺苷和甘草酸的含量测定。 Objective:To simultaneous determine the contents of cAMP and the glycyrrhizic acid in Xiaojianzhong granules by RP - HPLC. Methods: An external standard method by RP - HPLC with an Agilent TC - C18 (4. 6 mm × 150 mm, 5 μm)column at the temperature 30 ~C. The mobile phase consisted of methanol (A) - 0.01 mol .L^- 1 KH2 PQ ( B ) with the gradient elution (0 - 12 min, 90 % B ; 12 - 30 min, 30 % B ), at the flow rate of 1.0 mL .min^-1. The absorbance was monitored 250 nm. Results: Linearities of cMAP and glycyrrhizic acid were good in the ranges of 12. 5 -200 μg . mL^-1. The linear equations of cMAP and glycyrrhizic acid were Y = 23190 X - 4. 6083 ( R^2 = 0. 9998 ) and Y = 11364X + 80. 342 ( R^2 = 0. 9950 ). The average recoveries of cMAP and glycyr- rhizic acid were 95.2% and 97. 5% with the RSDs 2. 8% and 1.8%. Conclusions: The method is simple,believa- ble and can be used for determination of cAMP and glycyrrhizic acid in Chinese patent medicine xiaojianzhong granules.
