摘要
目的建立手性分离-液相色谱/质谱法,用于分离检测血液中艾司佐匹克隆。方法血液经HLB小柱固相萃取后,采用手性分离柱,用电喷雾正离子模式离子化、多反应监测模式检测艾司佐匹克隆,外标法定量。结果采用本文方法可有效分离佐匹克隆的左旋、右旋异构体,血中艾司佐匹克隆在1~100ng/mL范围内线性关系良好,相关系数为0.996 4,最低检出限为0.8ng/mL,回收率为76%~88%,日内与日间精密度均小于8%。结论本文所建方法简便、快速、分离度好,适用于血液中艾司佐匹克隆的检测。
Objective To establish a method of chiral separation-liquid chromatography/tandem mass spectrometry for determination of eszopiclone in blood. Methods blood sample was extracted using HLB solid phase extraction. The extract was separated on chiral separation column, confirmed and quantified by MS-MS in the multiple reaction monitoring (MRM) mode via positive electrospray ionization mode. Results Eszopiclone in blood was separated well. Calibration curves were linear within the range of 1 - 100ng/mL (R2 = 0. 996 4), and the limits of detection were 0. 8ng/mL. The recoveries were between 76% -88%, the inter-day and intra-day precision were less than 8%. Conclusion This method is fast and good resolution and suitable for determination of trace eszopiclone in blood.
