苯乙酸对胶质瘤细胞C6的C-myc基因蛋白水平表达的抑制作用及意义

摘要 目的 观察诱导分化剂苯乙酸对胶质瘤细胞C6的增殖分化过程中癌基因C-myc的蛋白水平变化。方法 〖^3H〗TdR掺入法测细胞增殖率,免疫组化SP法检测C-myc基因蛋白水平的表达。结果 应用苯乙酸后,细胞CPM降低;C-myc基因胞浆阳性表达显著下降。结论 苯乙酸对胶质瘤细胞存在剂量依赖性抑制作用机制可能为直接抑制胞浆中RNA合成。 Objective To determine the growth-inhibitory and differentiation-induing activity of phenylacetate (PA) on glioma cell C6. To find the mechanism of PA on molecule level.Methods The effect of PA on cell proliferation was deter-- mined by [~3H]-TdR assay; cell cycle distribution was analyzed with flow cytometry; and C-myc gene protein expression was studied by immunohistochemial technique.Results At doses of ranging 0.625,2.5,5.0mM, PA caused a dose-dependent prolif-- eration inhibitory effect. Cells arrested in Gall, Phases were observed in PA--treated (2.5.5.0mM) cells. The protein expres- sion of C--myc gene was notably decreased by PA (0. 625,2. 5mM). Conclusion PA caused a dose--dependent proliferation in - hibitory effect. One of the mechanisms of the effect should be PA decreased RNA amount directly in cells, although a lot of cells entered S phase, but most of them can not finish cell cycle. All of these effects connected with C-myc gene pro- tein expression were deceased by PA.
