摘要
目的 :观察肾性高血压 (RH)大鼠离体胸主动脉收缩反应变化并探讨其机制。方法 :将体重为 1 60~ 1 80 g雄性 Wistar大鼠随机分为高血压与对照(C)组 ,术后饲养 1个月后 ,用颈动脉摘管法测血压 ,用生物测定法观察胸主动脉收缩反应变化及 NO合成酶抑制剂 L -NAME及鸟苷酸环化酶抑制剂美蓝 (MB)对其的影响。用张力传感器记录血管环张力变化。结果 :RH组大鼠胸主动脉对苯肾上腺素 (PHE,1 0 - 6~ 1 0 - 9mol/ L )的收缩反应明显高于 C组 ,两组去内皮的血管环收缩反应相比无显著差异。加入 L-NAME及 MB后 ,两组收缩反应均明显增强 ,C组增加尤为显著。结论 :肾性高血压大鼠血管收缩反应增强 ,且与内皮有关。血管内皮通过
Objective:To observe the changes of thoracic aorta contraction of vasoconstriction in renal hypertensive rat and to study its mechanisms.Methods:Male Wistar rats weighing 160~180g were divided into two groups:the renal hypertensive (RH) group and the control (C) group.Blood pressures were measured by inserting a cannula into the carotid artery after feeding the rats for one month.Observe the changes of vasoconstriction and the effect of L-NAME(nitric oxide sythase inhibitor) or methylene blue (MB,guanylic acid cyclase inhibitor) by bioassay.Using force displacement transducers to record force changes.Results:The isometric contraction of the aortic rings in response to phenyle phrine(PHE,10 6 ~10 9 mol/L) was greater in RH rats than that of control ones.Removal of the endothelium made little difference in PHE-induced vasoconstriction between the two groups.The addition of L-NAME(10 4 mol/L) or MB(10 6 mol/L) increased the contraction response significantly in the both groups,but the effects were more pronounced in the control′s.Conclusion:The decrease of endothelium-derived relaxing factor(EDRF) systhesis and function might be related to the exaggerated vasoconstriction to PHE in aortic rings in RH rats.
