摘要
目的:测定猪血小板衍生生长因子的氨基酸组成和等电点。方法:用酸水解及聚丙烯酰胺凝胶等电聚焦的方法测定猪血小板衍生生长因子的氨基酸含量、组成成分和等电点。结果:酸水解法测定的猪血小板衍生生长因子含有17种不同的氨基酸,其中含有相对较多的碱性氨基酸,其等电点为9.8~10.3。结论:猪血小板衍生生长因子是一种强阳离子多肽,含有相对较多的碱性氨基酸,与其等电点为9.8~10.3是一致的。
Objective: To determine the amino acid composition and isoelectric point of porcine platelet-derived growth factor. Methods: In the present experiment, acid dehydroxy amino acid analysis and isoelectric focusing in polyacylatmide gels were used and the amino acid composition and isoelectric point of porcine platelet-derived growth factor were studied. Results: The findings of acid dehydroxy amino acid analysis and isoelectric focusing in polyacylamide gels showed that, porcine platelet-derived growth factor contained 17 amino acid residues in which the protein was rich in the strong predominance of basic amino acid and its isoelectric point was in the range of 9. 8-10. 3. Conclusion: Porcine platelet-derived growth factor is a strongly positive ionic polypeptide and may contain the strong predominance of basic amino acids in its amino acids composition, as reflected by the isoelectric point range of 9. 8-10. 3.
