摘要 探讨人参皂甙(GS)对人骨髓CD34+造血干细胞的增殖作用。方法:采给Dynal M450 CD34+免疫磁珠分离法阳性选择获取高纯度的人骨髓CS34+造血干细胞,应用多向祖细胞(CFU-Mix)体外培养技术,观察CD34+造血干细胞对GS的敏感性。结果:1.经Dynal M-450 CD34+免疫磁珠分离获得骨3髓 CD34+细胞占骨髓单个核细胞( MNC)的 0. 52~1.30%;CD34+细胞的存活率达94.0~98.2%。 2最佳浓度的GS使CD34+细胞的CFU-Mix集落形成率提高54.4%(P<0.01),而高浓度时表现为抑制。结论:GS不但作用于人骨髓造血祖细胞,而已对CD34+造血干细胞也具有明显的刺激增殖作用。 To investigate the proliferation effects of Ginsenosides (GS) on CD34positive hematopoietic stem cells from human bone marrow Methods: Highly Purified CD34+ hematopoietic stem cells were isolated from human bone marrow by using a Dynal CD34 Progenitor CellSelection System(Dynal Norway). The potentiated effects of GS on CD34+ hematopoietic stem cells were observed with an agarsemi - solid culture of CFU - Mix colony assay. Results: The number of CD34+ hematopoietic stem cells after being purified from Dynal beads system was 0. 52%- 1. 30%out of mononuclear cells of bone marrow Viable cells were 94. 0% - 98. 2%out of CD34positive cells GS could stimulate the proliferation of CD34+ hemetopoietic stem cells in colony culture assay, the numbers of CFU - Mix in the presence of GS 25and50mg/L were increased by 49. 3± 4. 9% and54. 4±6. 6% overthose of non - GS control respectively(P < 0. 01, each) Conclusion: The results above indicated that GS couldnot only stimulate the proliferation of progenitor cells, but also signiflcanty enhance the growgh ofCD34+ hematopoietic stem cells.
