摘要
建立高效液相色谱法同时测定丹参植株中不同部位丹参素、原儿茶酸、原儿茶醛、迷迭香酸和丹酚酸B五种水溶性成分的含量。采用DiamonsilTM C18色谱柱(250mm×4.6mm,5μm),以5%冰醋酸和甲醇为流动相进行梯度洗脱,柱温:35℃,流速为1.0mL/min,检测波长为288nm。丹参素、原儿茶酸、原儿茶醛、迷迭香酸和丹酚酸B的线性范围分别为0.35-21μg.mL-1(r=0.9999),0.3625-21.75μg.mL-1(r=0.9997),0.32-19.2μg.mL-1(r=0.9999),1.92-115.2μg.mL-1(r=0.9998),8.1-483.6μg.mL-1(r=0.9998)。平均回收率丹参素为99.3%(RSD 1.5%),原儿茶酸为99.7%(RSD 1.1%),原儿茶醛为99.8%(RSD1.7%),迷迭香酸为100.1%(RSD 0.4%),丹酚酸B为99.7%(RSD 1.5%)。本方法快速简便,5组分分离良好,检测结果为综合开发利用丹参地上部分资源提供科学依据。
To establish an HPLC with diode array detection for simultaneous determination of five watersoluble compounds: danshensu, protocatechuic aicd, pmtocatechuic aldehyde, rosmarinic acid and sal-vianic acid B in different part of Danshen. The column was Diamonsil^TM C18 (250mm×4.6mm,5um). The mobile phase was methanol and 5% acetic acid glacial with gradient elution and the column temperature was 35℃, the flow rate was 1.0mL/min, the detection wavelength was set up at 288nm. The linearity was obtained over the ranges of 0.35-21ug.mL-1 (r=0.9999) for danshensu with average recovery of 99.3%(RSD was 1.5%), 0.36-21.75ug.mL-1 (r=0.9997) for protocatechuic aicd with average recovery of 99.3%(RSD was 1.5%), 0.32-19.2ug.mL-1 (r=0.9999) for protocatechuic aldehyde with average recovery of 99.8% (RSD wasl.7%), 1.92-115.2ug.mL-1(r= 0.9998) for rosmarinic acid with average recovery of 100.1% (RSD was 0.4%) 8.1-483.6ug.mL-1 (r=0.9998) for sal-vianic acid B with average recovery of 99.7% (RSD wasl.5%). The method is rapid, simple, and reliable, and can be used for simultaneous determination of five watersoluble compounds in different part of Danshen.
