摘要
基于多成分对不同产地的丹参进行含量测定与质量评价。建立HPLC-UV检测方法,于波长210 nm处检测29批来自5个地区的丹参样品中以丹酚酸B、迷迭香酸为对照的酚酸类成分和以丹参酮IIA、隐丹参酮为对照的丹参酮类成分含量,进行质量评价和精密度、重复性、稳定性等方法学考查。SPSS 29.0进行系统聚类分析,SIMCA 14.1进行主成分分析和偏最小二乘法判别分析。迷迭香酸,丹酚酸B,丹参酮IIA,隐丹参酮分别在0.0255~1.2770,0.350~7.000,0.0115~0.2870,0.020~0.499的检测质量浓度范围内线性关系良好;精密度分别为0.92%,1.02%,0.90%,0.88%;重复性分别为1.90%,2.37%,2.95%,2.81%;稳定性分别为1.33%,1.39%,1.54%,1.62%;加样回收率分别为1.52%,1.22%,1.72%,1.41%。聚类分析结果表明,29批样品可聚为4类,其中P7单独为一类,P5、P14、P16-P17、P19-P21聚为一类,P1、P13、P18、P22-P26、P29聚为一类,P2-P4、P6、P8-P12、P15、P27-P28聚为一类;隐丹参酮和丹参酮IIA对于样品荷载值较大。偏最小二乘法判别分析结果表明,4种成分中,丹酚酸B、隐丹参酮、丹参酮IIA的VIP值>1.0。各产地丹参药材品质存在一定差异,以山东样品含量最佳。研究所建立的多指标含量测定方法简便、快速、准确性高、重复性好,可用于丹参的质量控制,为丹参的质量评价研究提供参考。
To determine the content and evaluate the quality of Salvia miltiorrhiza from different origins based on multiple components.An HPLC-UV detection method was established to measure the content of phenolic acids(using rosmarinic acid and salvianolic acid B as references)and tanshinones(using tanshinone IIA and cryptotanshinone as references)in 29 batches of Salvia miltiorrhiza samples from five regions.Quality evaluation was conducted,along with methodological investigations on precision,repeatability,and stability.Systematic cluster analysis was performed using SPSS 29.0,while principal component analysis and partial least squares discriminant analysis were carried out using SIMCA 14.1.Rosmarinic acid,salvianolic acid B,tanshinone IIA,and cryptotanshinone exhibited good linearity within the detection concentration ranges of 0.0255~1.2770,0.350~7.000,0.0115~0.2870,0.020~0.499,respectively.Precision values were 0.92%,1.02%,0.90%,and 0.88%,while repeatability values were 1.90%,2.37%,2.95%,and 2.81%.Stability values were 1.33%,1.39%,1.54%,and 1.62%,and the spiked recovery rates were 1.52%,1.22%,1.72%,and 1.41%,respectively.Cluster analysis revealed that the 29 batches could be divided into four groups:P7 formed a separate group,P5,P14,P16~P17,P19~P21 clustered together,P1,P13,P18,P22~P26,and P29 formed another group,and P2~P4,P6,P8~P12,P15,P27~P28 formed the final group.Cryptotanshinone and tanshinone IIA showed higher sample loadings.Partial least squares discriminant analysis indicated that among the four components,salvianolic acid B,cryptotanshinone,and tanshinone IIA had VIP values>1.0.There were significant differences in the quality of Salvia miltiorrhiza across different origins,with the samples from Shandong exhibiting the highest content.The multi-indicator content determination method established in this study was simple,rapid,highly accurate,and reproducible,making it suitable for quality control of Salvia miltiorrhiza and providing a reference for its quality research.
