摘要
目的研究肿瘤坏死因子基因TNF-α-308、TNF-α-238与广西肝癌家族聚集性的关系。方法在广西肝癌高发区选取12个肝癌高发家族(124例)及与之相对应的12个非肿瘤对照家族(129例)为研究对象,采用SNP芯片技术检测两组中TNF-α-308、TNF-α-238的基因型及等位基因频率。结果肝癌高发家族组TNF-α-308G/A基因型分布频率高于对照家族组,差异有统计学意义(22.6%vs11.6%,χ2=5.376,P=0.020);肝癌高发家族组TNF-α-308A等位基因分布频率亦高于对照家族组,差异有统计学意义(11.3%vs5.8%,χ2=4.877,P=0.024)。两组TNF-α-238基因型频率及等位基因频率差异均无统计学意义(P>0.05)。结论 TNF-α-308基因多态性可能与广西肝癌家族聚集性相关,携带TNF-α-308A等位基因可能会增加肝癌高发家族成员患肝癌的风险;TNF-α-238基因多态性与广西肝癌家族聚集无统计学关联。
Objective To explore the correlation between genetic polymorphisms of tumor necrosis factor (TNF-a-308,TNF-a-238) and family clustering of primary liver cancer(PLC ) in Guangxi. Methods Blood samples were collected from 124 families with a high incidence of PLC and from 129 control families.Genotype and alleles of TNF-a-308 and TNF-a-238 were determined by SNP microar- ray technology. Results The frequencies of TNF-a-308G/A genotype and TNF-a-308A allele in the PLC-clustering families (22.6% and 11.3% )were significantly higher than those in the control families (11.6% and 5.8% ).However,the two groups showed no statisti-cally significant differences in frequencies of the TNF-a-238 genotype or allele(P〉0.05). Conclusions TNF-a-308 genetic polymorph-ism is associated with susceptibility in families with high PLC incidence in Guangxi. In particular,the TNF-a-308A allele is proba-bly a risk factor for PLC in these families. In contrast,the TNF-a-238 polymorphism may not be associated with family clustering of PLC in Guangxi.
