摘要
应用荧光猝灭法、三维光谱法和圆二色谱法,研究了生理条件下芒果苷与胰蛋白酶的相互作用。结果表明:芒果苷对胰蛋白酶内源荧光的猝灭机制属于形成基态复合物所引起的静态猝灭。由van’t Hoff方程求得芒果苷与胰蛋白酶作用的热力学参数,焓变(ΔH)和熵变(ΔS)分别为70.45 kJ.mol-1和321.90 J.mol-1.K-1,表明芒果苷与胰蛋白酶之间的结合反应主要靠疏水作用驱动。根据Frster非辐射能量转移理论计算出芒果苷在胰蛋白酶中的结合位置与胰蛋白酶中色氨酸残基间的距离为3.99 nm。三维荧光光谱和圆二色谱显示,芒果苷诱导胰蛋白酶的构象发生部分改变。
The interaction of mangiferin with trypsin under physiological condition was studied using fluores- cence,three-dimensional fluorescence and circular diehroism spectroscopy. The results showed that the probable quenching mechanism of fluorescence of trypsin by mangiferin was a static quenching by forming the ground-state MA-trypsin complex. The thermodynamic parameters,enthalpy change (AH) and entropy change (AS) were calcu- lated to be 70.45 kJ ~ mo1-1 and 321.90 J ~ mo1-1 . K -1 via the van' t Hoff equation,which suggested that the hy- drophobic interactions plays major role in the binding of mangiferin to trypsin. The binding nm away from tryptophan residue in trypsin based on the F6rster theory of non-radioactive of three-dimensional fluorescence and CD spectra showed that the binding of tial changes in the secondary structure of the orotein. mangiferin to locality was an area 3.99 energy transfer.
