摘要
目的:建立血浆中黄酮哌酯活性代谢物MFCA浓度测定的HPLC法。方法:血浆样品经碱化、水解及酸化后,用乙酸乙酯提取。以甲醇-20mmol/LKH2PO4(70∶30)为流动相,紫外310nm检测。结果:线性范围0.25~32μg·ml-1,萃取回收率96.1%~100.3%。加样回收率96.8%~104.0%,日内RSD0.52%~2.30%,日间RSD1.39%~2.97%。结论:本法准确可靠。
OBJECTIVE:To establish a HPLC method for the determination of 3 methylflavone 8 carboxylic acid(MFCA),the main metabolite of flavoxate,in human plasma.METHODS:MFCA and the internal standard were extracted with ethyl acetate after alkali hydrolysis. The mobile phase consisted of methanol and 20 mmol·L -1 potassium dihydrogen phosphate(70∶30).The eluate was monitored at 310 nm.RESULTS:The calibration curve was linear within the range of 0.25~32 μg·ml 1 .The recoveries of extraction were 96.1%~100.3%.The recoveries of methodology were 96.8%~104.0%.Within day RSD ranged from 0.52% to 2.30%.Between day RSD ranged from 1.39% to 2.97%.CONCLUSION:This method is accurate and reliable.It has been applied successfully to the determination of MFCA in human plasma for the pharmacokinetic study of flavoxate.
