Development of stable isotope labeling technique and its application in encephalopathy research

摘要 寻找脑中特异的蛋白标记物,对于脑病临床诊断及药物机制研究至关重要。人脑是一个复杂的有机体,采用单靶点、非动态、局部的方法不适于脑病研究,而以质谱技术为基础的定量蛋白质组学的出现和发展,加快了脑病标志分子的研究进程。细胞培养稳定同位素标记技术(SILAC)作为精确的蛋白定量方法,是系统、动态的研究方法,为脑病机制和治疗脑病中药的研究提供了新的思路。 It is crucial to find out the brain specific protein markers for clinical diagnosis and drug mechanism.Human brain is a complex organism,so the single-target,non-dynamic and local method is not suitable for brain research.The development of quantitative proteomics,which is based on the mass spectrum,greatly accelerates the research process.As the accurate quantitative method of protein,the stable isotope labeling with amino acids in cell culture(SILAC) is a system,dynamic technology.It can provide new ideas for the mechanism research and the treatment strategy of traditional Chinese medicine in encephalopathy.
