前列腺素F2α对小鼠骨髓细胞CFU-GM增殖的双向调控作用

THE DUAL MODULATION EFFECT OF PROSTAGLANDIN F2α, ON CFU-GM PROLIFERATION OF MOUSE BONE MARROW IN VITRO

摘要 通过体外半固体琼脂培养分别观察PGF_(2α)对小鼠骨髓细胞CFU-GM增殖和骨髓细胞DNA合成的影响。测定结果表明,PGF_(2α)对CFU-GM及骨髓细胞具有双向调控作用。PGF_(2α)浓度在10^(-10)~10^(-8)mol/L时表现为刺激增殖作用,PGF_(aα)浓度在10^(-9)mol/L时刺激作用最强,集落形成比对照组多34.1%,~3H-TdR掺入率增加44.5%,而PGF_(2α)大于10^(-7)mol/L时则表现为抑制作用。集落形成与~3H-TdR掺入率的50%抑制率的PGF_(2α)浓度均接近于10^(-5)mol/L。同时,~3H-TdR掺入率的改变提示PGF_(2α)对CFU-GM增殖的调节是通过影响细胞内DNA合成来实现的。集落形态学分类结果表明,PGF_(2α)对粒系,巨噬系、粒单混合系三类型集落形成都表现为双向调控作用,而粒系对PGF_(2α)反应最为敏感。 The effect of prostaglandin F_(2α) (PGF_(2α)) on CFU-GM proliferation of the mouse bone marrow in vitro was evaluated in the present study. A dual effect of PGF_(2α) on mouse bone marrow CFU-GM was observed when PGF_(2α) was added to semisolid agar culture system or to suspension culture system with ~3H-TdR incorporation as the indicator. The proliferationpromoting effect occurred at PGF_(2α) doses ranging from 10^(-8) to 10^(-10) mol/L and the inhibitory effect appeared at PGF_(2α) doses ranging from 10^(-5) to 10^(-7) mol/L. The dose showing 50% inhibitory effect was found to be about 10^(-5) mol/L. Morphological analysis of mouse bone marrow hematopoietic cells indicated that CFU-G was more sensitive than CFU-GM and CFU-M to modulation effect of PGF_(2α). These findings suggest that PGF_(2α) plays an important role in modulation of hematopoietic stem cell growth.
