摘要
将构建好的可表达GST融合蛋白的重组病毒AcMNPV -OCC -Etp2 8感染Sf9细胞 ,72h后取感染了病毒的细胞裂解物上清液进行SDS -PAGE和Western -blot分析 ,结果显示 5 3kDa的融合蛋白 (GST -6xHis-Etp2 8)在昆虫Sf9细胞中得到了高效表达 ,表达水平为占细胞总蛋白的 2 1.3% ;将GST - 6xHis -Etp2 8注射艾维茵鸡进行免疫保护试验 ,结果表明免疫组比对照组在平均增重和盲肠粘膜层卵囊计数方面有一定程度改进 .
The recombinant viruses,AcMNPV OCC - GST 6xHis Etp28,obtained by co transfection,were used to transfect the Sf9 cells,and the supernatants of 72?h pi cell lysate was examined by SDS PAGE and Western blot.Results showed GST 6xHis Etp28 fusion protein of 53?kDa was expressed.Densitometric scanning of SDS PAGE gel revealed the expressed fusion protein accounted for 21.3% of the total cell proteins.GST 6xHis Etp28 was injected into chicken muscle to detect the immunoprotective effect of recombinant antigen.The results showed that immunized group had some improvement on wight increment and oocyst quantity of chicken cecal musosa.
