实验性衰老糖尿病大鼠模型研究 被引量:3

Study on experimental aged diabetes mellitus rats model

摘要 目的:进行人工诱导衰老糖尿病大鼠模型的初筛。方法:选用Wistar雌性大鼠,采用腹腔注射D-半乳糖后,给予不同周期高脂高糖饲料喂养,再配以不同剂量的链脲佐菌素(STZ)素腹腔注射,制备模型大鼠。检测空腹血糖(FBG)、超氧化物歧化酶(SOD)、丙二醛(MDA),并取胰腺组织行HE染色常规病理切片,以初筛实验性衰老糖尿病大鼠模型。结果:①各组成模率之间差异无显著性;②衰老糖尿病各组大鼠FBG升高,SOD下降,MDA升高;③胰腺病理切片显示:衰老糖尿病各组胰岛细胞团为正常对照组的22.93%-44.51%(P<0.01),且胰岛边界不清,岛内细胞排列不规则,细胞空泡化,细胞数目明显减少。结论:①不同造模法均可达到较高成模率。②采用腹腔注射D-半乳糖42d+高脂高糖6周+腹腔注射STZ(25mg/体质量.体表面积,2次)法所构建的实验性衰老糖尿病模型较为理想。 Objective: To elementary screen aging diabetes Mellitus (DM) rats model induced by artificial technology. Methods: After peritoneal injection with D-galactose, the Wistar female rats were feeded with different periodical enriched diet and intraperitoneal injected with different doses Streptozotocin (STZ). The bloods were collected to test fasting blood glucose (FBG), superoxide dismutase (SOD), maleic dialdehyde(MDA) and the issue of pancreatic gland was exteriorized to prepare pathological section to further observe. Results: (1) There were no significant differences of the modeling rate among all groups. (2) All aged DM Rats' FBG and MDA were raised, and SOD was down regulated. (3) The results of pancreas pathological section showed that pancreatic islet cell mass of all DM groups rats was 22.93%-44.51%(P〈0.01) compared with the normal control group, the bound of pancreatic islet was unclear, the arrangements of islet cells were unregulated, islet cells were alveolated, and islet cells quantity reduced markedly. Conclusion: (1) DM rats model could be induced by different artificial methods. (2) Aged DM rats model induced by which combined with D-galactose intraperitoneal injection for 42 days, enriched diet for 6 weeks and STZ intraperitoneal injection twice was the ideal one.
