摘要
目的 :观察体外原代培养的人牙乳头细胞内Smad2蛋白的表达 ,以及Smad2蛋白在转化生长因子 - β1(transforminggrowthfactor- β1,TGF - β1)信号转导中的作用。 方法 :原代培养人牙乳头细胞 ,用TGF -β1和骨形成蛋白 - 2 (bonemorphogeneticprotein ,BMP2 )刺激培养的细胞 ,免疫组化观察。结果 :TGF - β1和BMP2刺激组均可见Smad2蛋白表达 ,但与对照组相比无明显差异。TGF - β1组可见Smad2从胞浆转位至核内聚集 ,BMP2组未见此作用。结论 :人牙乳头细胞内存在Smad2蛋白的表达 ,Smad2能特异地转导TGF - β1信号至核内发挥作用 ,提示TGF - β1调控成牙本质细胞分化的作用可能是通过Smad2信号途径实现的。
AIM: To investigate the expression of Smad 2 protein in cultured human dental papilla cells (HDPC) and the role of Smad 2 protein in mediating transforming growth factor-β1(TGF-β1) signaling in the HDPC. METHODS: Immunohistochemical staining of HDPC derived from primary culture on stimulation from TGF-β1 or BMP2. RESULTS: The expression of protein of Smad 2 protein was found in HDPC on stimulation from TGF-β1 and BMP2, but no significant difference compared with control group. Smad 2 was found nuclear translocation accumulation of cytoplasmical localization, but BMP2 had no function. CONCLUSION: Smad 2 is present and activated by TGF-β1 in the HDPC. Smad2 may be function as a specific downstream molecule of TGF-β1 receptors mediated signal transduction pathway in the HDPC.
