摘要
为保护野生资源和实现栽培,以山野豌豆的嫩茎为材料,采用组织培养的方法,进行了愈伤组织诱导与分化,试管苗的生根和生根继代增殖培养,以及试管苗的移栽、定植的研究。结果证明:MS+6-BA0.4mg·L-1+2,4-D2.5mg·L-1是愈伤组织诱导培养的理想培养基;1/2MS+6-BA1.0mg·L-1+NAA0.1mg·L-1+GA31.0mg·L-1是愈伤组织分化培养的理想培养基;I/3MS+IAA0.1mg.L-’+ABTl.5mg.L-’是不定芽生根培养和试管苗生根继代增殖培养的理想培养基;试管苗生根继代增殖培养的繁殖速度为1株试管苗1年繁殖约1万株试管苗。试管苗移栽成活率为89.0%,定植成活率为97.3%;定植的试管苗保持了山野豌豆的所有植物学性状。
In order to preserve the wild resources and realize cultivation, by using tissue culture methods, the tender stems of Vicia amoena were used as materials to do the research on callus induction and differentiation, plantlets rooting and rooting of the subculture, and plantlet transplanting colonization. The results showed that MS+ 6-BA0.4mg·L-1+ 2,4-D2.5 mg·L-1 and 1/2MS+ 6-BA1.0 mg·L-1+ NAA0.1 mg·L-1+ GA31,0 mg·L-1 were the optimum media for callus induction and differentiation respectively. The ideal medium for adventitious buds rooting and rooting of the subculture was 1/3MS+ IAA 0.1 mg·L-1+ ABT1.5 mg·L-1. The propagation speed of tube seedlings could be about 10,000 per year. The survival rate of transplanted tube seedlings was 89.0% and colonization survival rate was 97.3%. The colonization of plantlets maintained all biological traits which wild plant has.
