摘要
目的评价抗p21Ras单克隆抗体KGH-R1、KGH-R2、KGH-R3与肝细胞肝癌(hepatocellular,HCC)及正常肝组织的免疫反应性,为该抗体的临床应用奠定基础。方法选取正常肝组织30例和HCC 30例,用本实验室前期构建的广谱抗p21Ras单克隆抗体KGH-R1、KGH-R2、KGH-R3进行免疫组化染色,计算各样本的阳性细胞百分率和组织学评分(HSCORE),比较各组免疫反应性。结果 KGH-R2、KGH-R3与HCC样本免疫反应的阳性率高于正常肝组织(P<0.05);KGH-R1、KGH-R2、KGH-R3与HCC和正常肝组织免疫反应阳性样本阳性细胞百分率、HSCORE分值比较差异均有统计学意义(P<0.05)。结论抗p21Ras单克隆抗体KGH-R1、KGH-R2、KGH-R3与HCC有较好的免疫反应性,与正常肝组织的免疫反应性很弱,可开发为HCC的治疗性抗体。
Objective To investigate the immunoreactivity of anti-p21Ras monoclonal antibody(mAb) KGH-R1,KGH-R2 and KGH-R3 in hepatocellular carcinomas and normal hepatic tissues,as well as to form the basis of antibody-based clinical applications.Methods Immunohistochemical staining was performed in 30 of hepatocellular carcinomas tissues and 30 of normal hepatic tissues using these three monoclonal antibodies prepared in our laboratory.The immunoreactivity of KGH-R1,KGH-R2 and KGH-R3 was evaluated in terms of percentage of positive cases,positive cells and histological scores(HSCORE).Results For both KGH-R2 and KGH-R3 mAb,there was a higher percentage of positive cases in hepatocellular carcinomas than in normal hepatic tissues(P0.05).All three antibodies,KGH-R1,KGH-R2 and KGH-R3,showed significantly a higher average percentage of positive cells and HSCORE in hepatocellular carcinomas than in normal hepatic tissues(P0.05).Conclusion Anti-p21Ras mAb KGH-R1,KGH-R2 and KGH-R3 have high immunoreactivity with hepatocellular carcinomas while only weak positive reaction with normal hepatic tissues.Therefore,they may be used as therapeutic antibodies for hepatocellular carcinomas.
