Aβ25-35对原代培养小鼠皮层神经元突触素和PSD-95表达的影响 被引量:2

摘要 目的:观察Aβ25-35对小鼠皮层神经元突触损伤作用,观察PSD-95和突触素的表达,进而探讨阿尔茨海默病的发病机制。方法:将小鼠(出生48h内)断头取脑,急性分离皮层神经元,在24孔培养基上培养7d,经NSE染色鉴定为神经元,将Aβ25-35配制成浓度为5μmol/L,分别在0h、1h、2h、4h、6h致伤皮层神经元(每个时间点分别为4孔,设对照组),经免疫组化观察突触素和PSD-95的表达情况。结果:Aβ25-35作用1h后突触素、PSD-95免疫反映复合物的灰度值明显下降,即二者的表达明显减少,1-6h与同期时间点对照组相比差异有显著性(P<0.01),于6h后突触素、PSD-95几乎未见到表达。
