摘要
目的评价不同浓度五倍子水提取物对牙科综合治疗椅水路生物膜的清除作用。方法根据干预所用的五倍子水提取物浓度分为A组与B组,两组在实验起始点形成相同的生物膜后进行干预。每天治疗结束后,A组将浓度为100mg/ml的五倍子水提取物、B组将浓度为62.5mg/ml的五倍子水提取物,分别抽吸入综合治疗椅水道内,并保留至次晨,连续使用4周。分别于干预前及干预后1、2、3、4周时采样,荧光黄染色后激光共聚焦电镜观察生物膜的形成情况,照片进行定量分析,同时采集手机冷却水培养计数细菌菌落数。结果生物膜荧光强度及细菌生长菌落数干预前两组比较,差异均无统计学意义(P>0.05);干预后同组不同时间点比较,差异均有统计学意义(P<0.05),组间比较,差异有统计学意义(P<0.05);浮游菌和荧光强度之间有相关性。结论中药五倍子水提取物能有效清除生物膜,清除效果与浓度、时间呈正比。
Objective To evaluate the clearing effect of different concentration Chinese Gallnut water extract on the biofilms in dental unit water lines. Methods The dental unit water lines were divided into two groups. The biofilms in the two groups were cleared by different concentration Chinese Gallnut water extract(100mg/ml vs. 62.5mg/ml) for four weeks. The formation of biofilms were observed through laser confocal microscopy after they were dyed by fluorescent yellow at the first,second, third and fourth week after intervention. At the same time,the samples were collected in the cooling water for bacteria culture. Results Before intervention,there was no significant difference in the fluorescent intensity or the bacteria colonies in the cooling water between the two groups(P〉0.05). After intervention,there were significant differences in the above index between the two groups(P〈0.05). There was positive correlation between the bacteria colonies and the fluorescent intensity. Conclusion Chinese Gallnut water extract can effectively remove the biofilm. The removal efficiency is proportional to the concentration and time.
