氨基葡萄糖对SD大鼠肝脏糖代谢相关蛋白表达差异性的研究

摘要 目的:研究氨基葡萄糖对SD大鼠肝脏中糖代谢相关蛋白表达的影响。方法:选取成年雌性SD大鼠48只,按体重随机分成4组,分别为氨基葡萄糖高、中、低剂量组和阴性对照组。分别灌胃500mg/Kg,250mg/Kg,125mg/Kg的氨基葡萄糖溶液和1ml/100g的生理盐水,连续灌胃28天。实验结束时处死试验动物,用免疫组织化学法检测肝脏中蛋白激酶,己糖激酶,一氧化氮合酶,葡萄糖转运蛋白4和葡萄糖转运蛋白2的表达情况。结果:试验期间各组动物生长发育情况良好,灌胃不同剂量氨基葡萄糖的大鼠肝脏中五种蛋白的表达与阴性对照组相比均无显著性差异(P≥0.05)。结论:氨基葡萄糖未导致肝脏中糖代谢相关蛋白表达的异常,提示服用氨基葡萄糖不会影响肝脏的糖代谢过程。 Objective:To research the effects of glucosamine to glucose metabolism in liver through a 28-day feeding study with SD rats.Methods: Forty-eight adult rats were randomly divided into four groups by their weights: one H group,one M group,one L group and one C group.All rats were oral administrated with either normal saline or GlcN via gavage.The H group,M group,L group were fed GlcN of 500mg/Kg,250mg/Kg,125mg/Kg respectively and the C group were fed normal saline of 1ml/100g every day.The rats were fed for 28 days.At the end of the 28th day,five hepatic physiological indices of these rats were measured by immunohistochemistry.Results: During the experiment,conditions of rats in each group were good.The rats of H,M and L group had normal hepatic physiological indices and no organ tissue damage was detected.Conclusion: From the results of this 28 days feeding test,we can conclude that feed different doses of GlcN to rats will not affect the metabolism of glucose in their liver.
