摘要
以自然受精的桑种子的下胚轴为材料进行组织培养 ,MS +NAA( 1 0mg) +6 BA( 0 .5mg)为培养基诱导的愈伤组织生长正常 ,具有不定芽的分化能力。MS +2 ,4 D( 1 .0mg/L) +6 BA( 0 .5mg/L)诱导的愈伤组织没有分化能力 ,结构疏松并呈现褐变现象。愈伤组织分化培养与继代培养中 ,内源激素测定发现 :培养前期 ,前者的ABA、IAA和GA的含量均较后者保持一个较高的水平 ,而培养后期 ,后者的ABA有较大的上升 。
Formation of adventitious bud from callus derived from the hypocotyl of Mulberry seeds has been completed successfully.Endogenous hormones content of calli was measured by ELISA.The calli induced by MS+NAA 1.0mg/L+BA 0.5mg/L grew vigorously and had the ability of differentiation.The content of IAA、ABA、and GA of which was higher than that of the calli,which didn′t have the ability of differentiation,induced by MS+2.4 D 1.0mg/L+BA 0.5mg/L.The IAA、GA and ABA levels in calli cultured on medium for differentiation were higher than that cultured on medium for subculture of callus during earlier culture stage.While the level of ABA of the latter calli was significantly higher during later stage,and the content of IAA and GA decreased rapidly.
