三氧化二砷对博菜霉素致肺纤维化大鼠肺组织的影响及机制探索

摘要 目的观察三氧化二砷对博莱霉素致肺纤维化大鼠的治疗作用,从细胞凋亡角度探讨其作用机理。方法192只SD大鼠随机分为4组:对照组、模型组、三氧化二砷组、地塞米松组,每组48只。根据造模后给药时间点,将各组分为2个亚组:造模后14天、28天干预组,每组24只。每个亚组根据给药时间的长短再分为给药14天、28天、56天3个小组,每组8只。观察项目:肺组织病理标本HE染色炎症和纤维化程度评分;免疫组化法检测凋亡相关基因C—myc的表达。结果(1)对照组肺泡炎和纤维化程度分级均较其余各实验组低(P〈0.05);除造模后28天开始干预的地塞米松28天组、56天组外,其余各干预组病理评分均较相应模型组低,但仅有造模后14天开始干预的三氧化二砷各组、地塞米松14天组有统计学意义。(2)所有药物干预组的C—myc表达量均显著高于相应模型组(P〈0.05)。结论适'-3剂量的三氧化二砷能减少博莱霉素致肺纤维化大鼠的肺部胶原纤维沉积,且致感染率显著低于地塞米松,早期应用疗效更佳。
