摘要
为获得胭脂花无菌苗,本试验利用单因子试验和正交试验设计,探索了不同消毒方法、基本培养基、大量元素浓度和蔗糖对无菌种子萌发和幼苗生长的影响。结果表明:种子表面消毒的最佳方法是以6%NaClO消毒6min;最佳无菌萌发基本培养基是MS或B5培养基;MS培养基大量元素浓度在1倍时,最适宜于胭脂花种子萌发;蔗糖含量在15~45 g/L范围内对发芽率的影响差异不明显。
In order to obtain aseptic seeding of Primula maximowiczii.The seeds sterilization method and the basic medium types,macronutrient element and sucrose concentration on aseptic seeding were investigated by single factor and orthogonal experiment design methods.The main results showed that,the best seed sterilization method was to be soaked in 6% NaClO for 6 min;the optimal basic medium for aseptic seeding was MS or B5;MS medium with full macronutrient element is suitable for seed germination;and there were no significant differences in the germination rates for all the sucrose concentration.
