酒精对对苯二胺引起家兔皮肤刺激性的影响 被引量:1

P-phenylenediamine Caused the Rabbit's Skin Irritation after Lavaging Alcohol

摘要 目的观察在酒精灌胃状态下,家兔背部皮肤涂抹对苯二胺后皮肤的刺激变化情况。方法将64只家兔随机分为四组,对照组即生理盐水灌胃组,其他三组分别为4mL/kg,6mL/kg,8mL/kg的酒精灌胃组,1次/d进行灌胃7d后,第8天在家兔背部给予6%对苯二胺溶液涂抹皮肤,对侧作自身对照,末次给药24h后洗去观察反应,根据《化妆品卫生规范》(2007年版)"皮肤刺激性/腐蚀性实验"皮肤刺激反应评分标准进行评分,并取受试侧皮肤做组织病理学检测。结果不同剂量酒精灌胃组家兔涂抹对苯二胺出现皮肤刺激体征,其程度随酒精灌胃量的增加有加重趋势,皮肤刺激平均积分增高,从0.25逐渐增高至2.0,根据刺激强度分级从无刺激,到有刺激,与对照组相比较,具有统计学意义(P<0.05);各酒精组间同时比较P<0.05,具有统计学意义。4mL/kg组组织病理学上无明显改变,6mL/kg组出现上皮变薄,8mL/kg组出现上皮改变,表皮水肿。结论酒精使对苯二胺的皮肤刺激性增强,使皮肤发生组织病理学改变。 Objective This study aims to observe the change of the rabbit's skin irritation caused by application of p-phenylenediamine on its back skin after lavaging alcohol. Methods A total of sixty-four rabbits were divided into four groups randomly, lavage with saline assigned to Group A, and lavage with different dosages of alcohol assigned to the other three groups. Thus, the rabbits were observed for seven days. Onthe eighth day, the liquor with 6% p-phenylenediamine was applied on one side of the rabbit' s back skin, and the other side of the back skin was taken as control.Based on the evaluating criterion of skin irritation reaction "Hygienic Standard for Cosmetics" (2007 version), the skin irritation experimental groups were observed for 24h after liquor application,and sampling of the skin was performed for histopathological ex-amination. Results The alcohol groups showed the increasing skin irritation reaction with increasing dose of alcohol, with scores between 0.25 to 2.0.According to stimulus intensity graded from no stimulation to stimulated, there was a statistical significance compared with the control group (P 〈 0.05).And among thealcohol groups' (P〈0.05).Histopathology was normal in 4mL/kg group. Epithelial thining in 6mL/kg group. Epithelial edema in 8mL/kg group. Conclusion The alcohol is able to enhance the irritant effect of the p- phenylenediamine which is the main component of hair dye.
