摘要
羧肽酶E(Carboxypeptidase E,CPE)是多种激素肽和神经递质生物合成过程中最主要的羧肽酶.GEO数据分析显示,羧肽酶E作为激素原处理酶,在不同类型的肿瘤中表达.在肺神经内分泌肿瘤、垂体瘤等中,CPE mRNA表达升高.微阵列分析显示,在很多转移性非神经内分泌肿瘤中,CPE mRNA的表达水平也显著升高,例如:结直肠癌,肾癌,尤文氏肉瘤,但CPE mRNA的表达在其相应的正常组织中缺如.研究显示,不仅野生型CPE在肿瘤中表达,其变异体也与肿瘤相关,在一些较大或侵袭性肿瘤中,CPE 变异体mRNA的基因拷贝数非常高.总之,CPE可能在促进肿瘤生长、侵袭及转移中发挥重要作用,能够成为不同类型转移癌诊断及预后判断潜在的生物标志物.
Carboxypeptidase E (CPE)is a key carboxypeptidase, and it is involved in the biosynthesis of peptide hormones and neurotransmitters. CPE as a prohormone processing enzyme expressed in different cancer types was analyzed the data in the GEO profile database. Elevated CPE mRNA expression was found in neuroen- docrine tumors in lung and pituitary adenomas, Microarray studies demonstrated that significantly elevated levels of CPE mRNA was found in many metastatic non - endocrine cancers ( colon rectal, renal cancers and Ewing sarcomas) ,whereas expression of CPE mRNA was absent in their respective counterpart normal tissues. Not only wild - type CPE was expressed in tumors, but a variant was correlated with tumor behavior. In very large or inva- sive tumors,the copy numbers of the variant of CPE mRNA were very high. On the whole,CPE may play an im- portant role in promoting tumor growth, invasion and metastasis. CPE could be used potentially as a diagnostic and prognostic biomarker for metastasis in different cancer types.
