低分子量肝素对SAP大鼠肾组织保护的实验研究 被引量:5

摘要 目的观察低分子量肝素(LMWH)对大鼠重症急性胰腺炎(SAP)肾组织热休克蛋白70(HSP70)表达、肾组织的光镜下形态学的改变及血清尿素氮(BUN)和肌酐(Cr)水平的变化,探讨LMWH对肾组织保护的作用机制。方法选用Wistar大鼠随机分为3组,假手术组(A组)、SAP模型组(B组)、SAP低分子量肝素治疗组(C组),术后24h和72h分别取肾组织,HE染色检测肾组织形态学变化,免疫组化检测肾组织HSP70的表达,并测定大鼠血清BUN和Cr。所有实验数据均采用均数±标准差(x±s)表示,统计学处理用F检验。结果肾组织HSP 70表达:各组肾组织HSP70表达在术后24h比72h多,统计学处理有显著性差异(P<0.05,P<0.01)。A组HSP70表达较少,B组HSP70表达最多,C组HSP70表达明显下调,与B组比较有显著性差异(P<0.01)。光镜下可见:A组大鼠双侧肾脏无明显病理改变;B组肾小管上皮细胞坏死脱落,肾小管水肿;C组肾小管水肿程度较B组明显减轻,未见肾小管上皮细胞坏死,但有红细胞渗出。血清Cr和BUN含量:C组与B组比较,有显著性差异(P<0.01,P<0.015)。结论 LMWH通过下调肾组织内HSP70表达,减轻SAP大鼠肾的损伤,从而起到保护肾组织的作用。 【Objective】 To observe the protective effect of low molecular weight heparin(LMWH) in rats with severe acute pancreatitis(SAP) on renal injury heat shock protein 70(HSP70) expression,kidney tissue optical microscope the morphology of the change and serum urea nitrogen(BUN),creatinine(Cr) level changed,this paper discusses on the protection of the renal tissues LMWH potential mechanism.【Methods】 Choose Wistar rat randomly divided into three groups,sham-operate group(group A),SAP model group(group B),SAP low molecular weight heparin treatment group(group C),postoperative 24h and 72h renal tissues were taken,HE dyeing detection kidney tissues morphological changes,immunohistochemical method detection kidney tissues HSP70 expression,with to measure rat serum BUN and Cr.All the experimental data are used mean ± standard deviation(plus or minus) said,statistics processing in Finspection.【Results】 Renal tissue's HSP70: The expression of HSP70 in each group at 24h was more than 72h.Statistics indicates a distinctive difference(P0.05,P0.01);group A HSP70 express less,groupB HSP70 express the most,HSP70 groups C express clear cut,and group B comparison,significant difference(P 0.01).By Light Microscopy: group A rats,bilateral renal tubular no significant pathological changes;group B of renal tubular epithelial cell necrosis fall off,renal tubular edema;groups C tubular edema degree is group B significantly reduce,did not see the tubular epithelial cell necrosis,but there are red blood cells diapedesis.Serum Cr and BUN content: groups C and group compared with B,significant difference(P 0.01,P0.05).【Conclusion】 LMWH cut through the renal tissues HSP70 expression,reduce rat renal damage SAP,which have protection kidney the function of the organization.
