摘要
目的研究6,8-二-三氟甲基-5-羟基-7-乙酰氧基白杨素(dFMAChR)的体外抗胃癌作用。方法体外培养人胃癌SGC-7901细胞。MTT比色法测定细胞活性。PI染色流式细胞术(FCM)分析细胞凋亡率。WesternBlot检测细胞NF-κB、Bcl-2、Bax蛋白表达。结果 dFMAChR显著抑制体外培养人胃腺癌SGC-7901细胞活性,呈剂量依赖性;dFMAChR有效诱导人胃腺癌SGC-7901细胞凋亡;dFMAChR下调NF-κB(p65)、Bcl-2蛋白表达,同时,上调Bax蛋白表达,呈浓度依赖性。结论 6,8-二-三氟甲基-5-羟基-7-乙酰氧基白杨素具有抗胃癌作用,其机制可能与其抑制NF-κB活化、下调Bcl-2蛋白表达和上调Bax蛋白表达相关。
Objective To investigate the antitumor effect of 6,8-ditrifluoromethy-5-hydroxyl-7-acetoychrysin(dFMAChR)on human gastric cancer cells cultured in vitro.Method SGC-7901 cells were cultured in vitro.MTT assay was used to test cell viability.PI staining flow cytometry(FCM)was used to analyze the apoptosis rate.The levels of NF-κB,Bcl-2 and Bax protein expression were examined by Western blot.Results dFMAChR significantly inhibited cell viability of SGC-7901 cells and induced apoptosis,in a dose-dependent manner.Westernblotting analysis indicated the levels of NF-κB,Bcl-2 protein expression were decreased and the level of Bax protein expression was increased,in a concentration-dependent manner after treatment with dFMAChR.Conclusions dFMAChR possess the antitumor effect against human gastric cancer and which underlying the mechanism of this action maybe mediated by down-regulation of NF-κB,Bcl-2 expression and up-regulation of Bax expression.
