以苯胺为原料 ,经酰化、环化、苯甲酰化 ,制得 5 - (苯甲酰基 ) - 3-甲基苯 -吲哚酮 (2 ) ,化合物 (2 )在氢氧化钡溶液中开环、重氮化、次磷酸还原 ,一锅制得酮基布洛芬 (1) ,总收率 31.1%
Aniline was used via amidation、cyclization、Friedel-Crafts reaction to give 5-benzoyl-3-methylindolin-2-one(4).(4)was hydrolysed in the presence of Barium hydroxide,then diazotizated、reduced by H 3PO 2 to afford ketoprofen with an overall yield of 311%.
