摘要
目的建立高效液相色谱法-二极管阵列法(HPLC-DAD)测定中药夜关门中牡荆苷含量的方法。方法采用岛津VP-ODS(4.6mm×150mm,5μm)色谱柱;以甲醇-0.3%冰醋酸溶液为流动相进行梯度洗脱;DAD检测器200～400nm波段扫描,338nm定量;柱温:30℃;流速:1mL/min;进样量:5μL。结果牡荆苷在49.1～491.0μg/mL范围内线性关系良好,平均加样回收率为97.38%,RSD=2.0%。7批不同来源样品中牡荆苷的平均含量分别为0.287%、0.354%、0.387%、0.231%、0.254%、0.176%、0.199%。结论所建立的含量测定方法简便、准确,可作为夜关门中牡荆苷的含量测定方法。
Objective To establish a HPLC-DAD method for determination of vitexin in Lespedeza cuneata.Methods The separation was achieved on a SHIMDAZU VP-ODS column（4.6 mm×150 mm,5 μm） with a gradient elution of methanol-0.3% acetic acid as the mobile phase.DAD detector 200 to 400 nm band scanning,338 nm quantitative.The column temperature was maintained at 30 ℃.The flow rate was 1 mL/min.Into the sample weight：5 μL.Results The linear range of vitexin was 49.1-491.0 μg/mL.The average recovery was 97.38%,RSD=2.0%.The average content of vitexin in 7 samples from different sources was 0.287%,0.354%,0.387%,0.231%,0.254%,0.176%,0.199%.Conclusion The method is accurate and repeatable,which is better for quality evaluation of vitexin in Lespedeza cuneata（Dum.Cours.） G.Don.
