肾移植稳定病人淋巴细胞表面淋巴细胞功能相关抗原-1、L-选择素的表达 被引量:5

Dynamic changes in CDlla, CD62L expression in lymphocytes in renal allograft recipients with stable course

摘要 目的 研究尸肾移植后稳定病人淋巴细胞表面淋巴细胞功能相关抗原 1(CD11a)、L 选择素(CD62L)表达的动态变化。方法 利用单克隆抗体 流式细胞仪荧光免疫技术 ,测定 10例肾移植后稳定病人不同时间外周血淋巴细胞CD11a、CD62L抗原表达 ,并以 10例移植前病人对照。结果 肾移植前组CD11a、CD62L抗原表达率分别为(77.3±16.3 ) %和(3 8.5±16.8) %。移植后 3、6、9、15d时CD11a抗原表达率分别为 (2 7.9±9.2 ) %、(2 8.5±7.7) %、(2 2 .9± 8.9) %和(5 6.2±17.1) % ,均低于术前组 (P <0 .0 5 ) ;CD62L分别为(3 1.5±14 .5 ) %、(3 3 .5± 12 .8) %、(42 .0±15 .3 ) %和(5 3 .8±15 .7) % ,与术前组比较差异无显著性 (P>0 .0 5 )。结论 淋巴细胞CD11a、CD62L表达水平和肾移植病人免疫状态密切相关。免疫抑制剂能明显抑制淋巴细胞CD11a表达 ,这可能是其预防早期排斥反应的重要机制之一。 Objective\ To investigate the dynamic changes in CDlla and CD62L on lymphocyte from peripheral blood of renal allograft recipients with stable course. Methods\ The dynamic changes in CDlla and CD62L levels on lymphocytes were examined by using mAb FACS immunofluorescence technique in 10 renal allograft recipients with no rejection within 15 days postoperatively. Ten patients undergoing chronic hemodialysis program were inspected simultaneously as control group. Results\ The expression of CDlla and CD62L on lymphocytes in control group were (77.3±16.3)% and (38.5±16.8 )% correspondingly. For stable course at the 3rd, 6th, 9th and 15th day after operation, the CDlla levels were (27.9±9.2)% ( P <0.001), (28.5± 7.7)% ( P <0.001), (22.9±8.9)% ( P <0.001), (56.2±17.1)% ( P <0.05) correspondingly, which were significantly lower than in the control group. The CD62L levels were (31.5± 14.5 )%, (33.5±12.8)%, (42.0±15.3)%, (53.8±15.7)% respectively without significant difference between the two groups ( P >0.05). The expression of CD62L was positively or negatively correlated to the platelet and the blood urea nitrogen correspondingly. Conclusion\ The expression levels of CDlla and CD62L on lymphocytes are closely correlated with the renal allograft recipients immune status. The immunosuppressants, especially methylprednisolone, can suppress the expression of CDlla on lymphocytes evidently, which may be one of the most important mechanisms in protection from the early graft rejection; and ATG may play important roles in keeping the lower level of CDlla and causing the induction of immunologic tolerance.
