摘要 目的探讨SFRP1基因甲基化及其蛋白在非小细胞肺癌组织中的表达情况。方法用免疫组化SP法和甲基化特异性PCR法检测60例非小细胞肺癌组织,21例肺良性疾病肺组织中SFRP1基因甲基化及SFRP1蛋白的表达情况。结果肺癌组织SFRP1基因甲基化阳性高于肺良性疾病肺组织,差异有显著性(P=0.001);SFRP1基因甲基化与非小细胞肺癌细胞分化程度及淋巴结转移情况有明显的相关性(P<0.05);非小细胞肺癌中SFRP1蛋白阳性率低于肺良性疾病肺组织,差异有显著性(P=0.004);SFRP1蛋白表达与非小细胞肺癌临床分期、细胞分化程度及淋巴结转移情况有明显的相关性(P<0.05);SFRP1甲基化和SFRP1蛋白表达缺失存在相关性(P=0.002)。结论 SFRP1基因甲基化可能是导致SFRP1蛋白表达降低的重要机制,进而导致非小细胞肺癌的发生发展。 Objective To investigate the expression of SFRP1 gene methylation and protein in non-small cell lung cancer.Methods The SP immunohistochemistry and methylation-specific PCR were applied to detect the SFRP1 methylation and protein in tissues of 60 cases of non-small cell lung cancer and 21 cases of lung benign diseases.Results SFRP1 gene methylation rate in non-small cell lung cancer was significantly higher than that in normal lung tissues(P=0.001).The loss rate of SFRP1 protein expression in non-small cell lung cancer was significantly higher than that in normal lung tissues(P=0.004).SFRP1 gene methylation was closely correlated with lymph node metastasis and degree of differentiation in non-small cell lung cancer(P0.05).SFRP1 gene protein expression was correlated with clinical stage,degree of differentiation and lymph node metastasis in non-small cell lung cancer(P0.05).SFRP1 gene methylation was closely correleted with to SFRP1 gene protein expression in non-small cell lung cancer(P=0.002).Conclusions Expression of SFRP1 gene methylation and protein is associated with the carcinogenesis and development of non-small cell lung cancer.
