钌红、丁卡因对肺动脉高压模型大鼠肺动脉平滑肌细胞[Ca^(2+)]i释放功能的影响 被引量:1

摘要 目的:分析肺动脉高压时肺动脉平滑肌细胞雷诺定(Ryanodine)受体[Ca2+]i释放功能的改变及钌红(Rutheniumred)、丁卡因对肺动脉高压模型大鼠肺动脉平滑肌细胞[Ca2+]i释放功能的影响。方法:清洁级Wister大白鼠25只,腹腔注射野百合碱,建立大鼠肺动脉高压模型,死亡2只,最终23只;原代培养肺动脉平滑肌细胞;Fura-2/AM(钙离子荧光指示剂)负载培养细胞;荧光测钙技术比较雷诺定诱导的野百合碱组(n=15)及对照组(n=8)[Ca2+]i数值,观测钌红、丁卡因对雷诺定受体激动剂雷诺定诱导的[Ca2+]i变化的影响。结果:10 nmol/L雷诺定使对照组[Ca2+]i平均增加(93.31±12.41)nmol/L,使野百合碱组[Ca2+]i平均增加(141.71±13.59)nmol/L;两组样本[Ca2+]i增加的数值比较差异有统计学意义(P<0.01)。钌红浓度为0.5~10μmol/L、丁卡因浓度为2~40μmol/L时,可使雷诺定受体激动剂雷诺定诱导的[Ca2+]i值最大下降(115.32±7.47)nmol/L和(107.97±7.11)nmol/L。结论:肺动脉高压大鼠肺动脉平滑肌细胞对雷诺定受体激动剂雷诺定的敏感性增强;钌红、丁卡因对雷诺定诱导的肺动脉高压大鼠肺动脉平滑肌细胞[Ca2+]i上升有明显的抑制作用。 Objective: To analyze the effect of ruthenium red and tetracain for ryanodine induced [Ca^(2+)]i releasing in pulmonary artery smooth muscle cell(PASMC) in rats with pulmonary artery hypertension(PAH). Metnods: The experimental rats were divided into 2 groups.PAH group,n=15,the rats received intraperitoneal injection of monocrotaline 60 mg/kg,and Control group,n=8,the rats received the same volume of normal saline.PASMC was primarily cultured with fluorescence Fura-2/AM.The effect of ryanodine,ruthenium red and tetracaine for [Ca^(2+)][Ca^(2+)]i releasing in PASMC were measured by fluorescence microscopy. Results: With 10 nmol/L ryanodine treatment,the [Ca^(2+)]i releasing in Control group increased by the average of(93.31±12.41) nmol/L,in PAH group increased by the average of(141.71±13.59) nmol/L,P0.01 respectively.With(0.5~10) μmol/L ruthenium red treatment,and with(2~40)μmol/L tetracaine treatment,in PAH group,ryanodine induced i releasing decreased at the maximum as(115.32±7.47)nmol/L and(107.97±7.11) nmol/L. Conclusion: The PASMC in PAH rat has the higher sensitivity to ryanodine receptor agonist.Ruthenium red and tetracaine can inhibite the ryanodine induced PASMC [Ca^(2+)]i releasing in PAH rats.
