虎金颗粒对大鼠肝纤维化的作用及其对肝脏AngⅡ含量及其受体AT1的影响 被引量:1

EFFECTS OF HUJIN GRANULE ON RATS’LIVER FIBROSIS AND ANG II AND AT1 RECEPTOR CONTENTS IN LIVERS

摘要 目的:观察虎金颗粒的抗肝纤维化作用和对肝纤维化模型大鼠肝脏组织血管紧张素Ⅱ(AngⅡ)含量及其1型受体(AT11R)表达的影响。方法:采用二甲基亚硝胺(DMN)腹腔注射诱导肝纤维化模型,实验分为模型组、大黄虫丸组及虎金颗粒低中高剂量组,另设正常组作对照。给药4周后,检测大鼠肝功、血清肝纤维化指标(HA、LN、PCⅢ)及肝组织AngⅡ、羟脯氨酸(Hyp)含量;采用免疫组织化学SABC法测定肝组织AT1R表达的变化。结果:虎金颗粒能改善大鼠肝功能,降低肝纤维化指标血清透明质酸(HA)、层粘蛋白(LN)、Ⅲ型前胶原(PCⅢ)的含量和肝组织Hyp的含量,与模型组比较有明显差异(P<0.05,P<10.01);降低大鼠肝组织AngⅡ含量(P<0.05,P<0.01);抑制大鼠肝组织AT1R的表达。结论:虎金颗粒有较好的抗肝纤维化作用,其作用机理与降低肝组织AngⅡ含量、抑制AT11R表达有关。 Objective: To observe the effects of hujin granule on rats’liver fibrosis and angiotension II( AngⅡ) content and angiotension II type 1receptor ( AT1R) expression in hepatic tissue. Methods: Models with hepatic fibrosis were induced by interaperitoneal injecting dimethylnitrosamine ( DMN) ,rats were divided into model,daihuang zhechong pill,hujin granule low,middle and high dose groups randomly,normal rats were taken as control,each group was given the corresponding medicine for 4 weeks. The hepatic function indexes were determined by using full automatic bio- chemical analyzer,serum hepatic fibrosis indexes such as HA,LN an PC III were examined by radioimmunassay,Ang II content in hepatic tissue were detected by radioimmunassay,Hyp content in hepatic tissue was determined by digestion method,the angiotension II type 1 receptor ( AT1R) expression in hepatic tissue was determined by immunohistochemistry method. Results: To compare with model group,hujin granule could significantly improve hepatic function of rats with liver fibrosis,lower ser- um HA,LN and PC III contents,decrease Hyp content in hepatic tissue of rats; hujin granule could obviously reduce Ang II content in hepatic tissue,inhibit the expression of AT1R in hepatic tissue. Conclusion: Hujin granule has good anti hepatic fibrosis effect,its mechanism was related to decreasing Ang II content in hepatic tis- sue and inhibiting AT1R expression.
