摘要
目的:探索建立将经典的电生理监测与目前比较前沿的小动物活体成像技术联用的实验方案,评价其在电针效应机制研究中的应用性。方法:运用电生理技术观察不同强度的电针刺激郄门穴对心动过缓模型大鼠心率(HR)、平均动脉压(MAP)、左心室收缩压(LVSP)的影响;采用Cy7荧光染料标记心得安,裸鼠注射后进行荧光活体成像检测,对其药物分布情况进行实时摄取图像。结果:不同的电针强度对于大鼠心动过缓调节存在效应差异,对HR、LVSP的影响,均为6mA>4mA>2mA。裸鼠体内活体成像,各器官能呈现比较好的药物浓度代谢规律,随着电针强度的增加,体现出药物排泄加快的趋势。结论:电生理结果与活体成像结果趋于一致,该实验方案对于从不同角度研究针灸的效应机制具有一定价值。
Objective: To explore the establishment of the experimental scheme of the classical electrophysiological monitoring combined with the current outlying small animals living imaging technology to evaluate its application on the study of mechanism of EA effect.Methods: To observe the effects of heart rate(HR),mean arterial pressure(MAP),left ventricular systolic pressure(LVSP) by stimulating Ximen point of bradycardia rat models by EA with different strength(2mA,4mA,6mA) using electrophysiological technique;to label propranolol using fluorescent dye Cy7,taking fluorescence imaging detection of naked mice after they have been injected,taking real-time images of their drug distribution.Results: There are differences in the effects of using different strength of EA in adjusting bradycardia of rats,the effects of heart rate,left ventricular systolic pressure are 6mA 4mA 2mA.naked mice in vivo imaging,each organ can present better concentration metabolic regularity.With increasing of EA intensity,it reflecting the accelerating trend of drug excretion.Conclusion: The electrophysiological results and the results of fluorescence imaging detection are consistent;this experimental scheme is feasible,with a sense of exploration and innovation,having an important application value for in-depth study the effects of acupuncture mechanisms from different points of view.
