摘要 目的探讨血管内皮细胞对血管平滑肌细胞增殖和分泌的的影响。方法血管平滑肌细胞接种于培养板中分为3组:TNF-α刺激组、无TNF-α刺激组和对照组,前两组分加入含有或不含TNF-α刺激的内皮细胞培养液,对照组不添加内皮细胞培养液。ELISA法测定初始和培养24h后血管平滑肌细胞培养液中ICAM-1、IL-8、IL-6水平,MTT法检测血管平滑肌细胞增殖情况。结果研究结束时,3组的ICAM-1和IL-8均较基线值有所增加,其中刺激组ICAM-1和IL-8较基线升高程度最高,均显著高于对照组(P<0.05)。3组平滑肌细胞的MTT值依次为(1.35±0.51)、(1.01±0.31)和(0.29±0.19),有显著性差异(P<0.05)。3组平滑肌细胞MTT值与培养液中的IL-6水平呈正相关(r=0.668,P<0.05)。结论 TNF-α诱导内皮细胞活化并释放IL-6,活化的内皮细胞能够促进血管平滑肌细胞增殖和细胞因子的释放。 Objective To investigate The influence of endothelial cells on the proliferation and secretion of vascular smooth muscle cells. Methods The vascular smooth muscle cells were inoculated on cultivation plates and divided to 3 groups as follows, TNF-α-activated group, the inactivated group and the control group cultured with or without the supernatant fluid of endothelial cell with or without TNF-α stimulation. All the supernatant fluid was collected at the 0h and 24h after grouping, then ELISA was used to measure the level of ICAM-1、IL-8、IL-6. The proliferation of the vascular smooth muscle cells were detected by MTT. Results At the end of the experiment, ICAM-1and IL-8 were increased from baseline value in all groups, but the maximum changes ((0.22±0.068)ug/ml and(2.78±0.957)ng/ml) were observed in Activated group. The MTT of the 3 groups were(1.35±0.51)、(1.01±0.31) and (0.29±0.19) with significant difference(P0.05). There was a significant correlation between MTT and IL-6 in supernatant fluid in 3 groups. Conclusion The proliferation and the cytokine release of the vascular smooth muscle cells were promoted by TNF-α activated endothelial cells through elevated the level of IL-6.
