摘要
丙酰螺旋霉素(Pro—SPM)是药生所利用酶工程定向酰化螺旋霉素(SPM)得到的一种新的十六碳大环内酯类抗生素,体内外抗菌活性优于临床目前使用的乙酰螺旋霉素(Ac—SPM)。本文选用μ—Bonadapack C_(18)柱,以甲醇—磷酸(PH2.8)(33:67)为流动相,较好地分离了兔血尿中Pro—SPM、Ac—SPM和SPM,并用单组份标准品建立了高压液相色谱法(HPLC)测定免血尿中Pro一SPM、Ac—SPM和SPM浓度的标准曲线,线性范围为0.1~8μg/ml。
Propionylspiramycin (Pro-SPM) is a new 16-mem-bered macrolide antibiotic by acylating spiramycin (SPM) directly with technologof enzyme by IMB. which is more active in vivo and in vitro than Acety-lspiramycin (Ac-SPM) used in clinical. Pro-SPM, Ac-SPM and SPM in rabbit plasma and urine were separatedby using μ-Bondapak C_(18) column with a mobile phase composed of Methylalcohol-Phosphoric acid (pH2. 8) (33: 67 ). Standard curves measuring theconcentrations of Pro-SPM, Ac-SPM and SPM in rabbit plasma and urinewere estabolished with the single standard components of Pro-SPM, Ac-SPM and SPM. Linear range was 0. 1-8. 0μg/ml.
