摘要
建立了高效液相色谱-荧光检测法测定水产品中3种四环素类药物(四环素、土霉素、金霉素)残留量的方法。均质后的样品用Na2EDTA-柠檬酸缓冲溶液提取,经HLB固相萃取柱净化,反相色谱柱分离,荧光检测器检测后外标法定量。3种四环素类药物的线性范围为0.05~10.0 mg/kg,线性相关系数均大于0.9990。样品在0.05、0.5、5.0 mg/kg 3个添加水平下的平均回收率为84.0%~91.3%,相对标准偏差为3.89%~8.65%。四环素、土霉素的方法检出限为0.01 mg/kg,金霉素为0.02 mg/kg。本方法适用于水产品中三种四环素类抗生素残留的同时确证检测。
A method for determination of 3 tetracyclines residues(tetracycline,oxytetracycline,chlortetracycline) in aquatic products using high performance liquid chromatography(HPLC) equipped with fluorescence detector was established.The homogenized sample was extracted with Na2EDTA-citric acid buffer solution,purified by Oasis HLB solid phase extraction column,separated on a RP-UPLC column and then detected by fluorescence detection.Quantitation was carried out using the external standard.The standard curves for all the compounds were linear over the concentration range of 0.05 to 10.0 mg/kg with correlation coefficients all above 0.999.The spiked recoveries at three levels of 0.05,0.5,5.0 mg/kg were ranged from 84.0% to 91.3%,and the relatives standard deviations(RSD) were in the ranges of 3.89% to 8.65%.The limits of detection were 0.008 mg/kg for tetracycline,and oxytetracycline,and 0.016 mg/kg for chlortetracycline.This method was suitable for simultaneously confirm of three tetracyclines in aquatic products.
