摘要
目的研究棕榈酸对大鼠主动脉内皮细胞凋亡的影响。方法 0.5%BSA或0.5%BSA及不同浓度的棕榈酸(0.1、0.2和0.3 m m)作用于大鼠主动脉内皮细胞24 h,M TT法检测细胞活性,流式细胞仪AnnexinⅤ/PI法检测细胞凋亡,分光光度计检测C aspase-3活性。结果 M TT检测结果提示棕榈酸组较对照组细胞活性下降;AnnexinⅤ/PI法检测结果提示棕榈酸组较对照组细胞凋亡增加(P<0.05或P<0.01)。棕榈酸组较对照组C aspase-3活性增加(P<0.05或P<0.01)。结论棕榈酸能够诱导内皮细胞凋亡,说明高水平的游离脂肪酸对内皮细胞有直接的伤害作用。
【Objective】We investigated the effect of ghrelin on palmitate-induced apoptosis in rat aortic endothelial cells.【Methods】Rat aortic endothelial cells were incubated in 0.5% BSA or 0.5% BSA complexed to palmitate(0.1 mm,0.2 mm,0.3 mm).Cell viability was assessed by MTT assay.Apoptosis was detected using an annexin V/PI double staining assay.Spectrofluorometer assay caspase-3 activity.【Results】Palmitate decreased cell viability compared with control group as assessed by MTT asssy.Palmitate increased cell apoptosis compared with BSA group as assessed by an annexin V/PI double staining assay(P 0.05 or P 0.01).Palmitate increased caspase-3 activity compared with BSA group(P 0.05 or P 0.01).【Conclusions】These findings provide the evidence that palmitate could induce apoptosis in rat aortic endothelial cells.High level of free fatty acid could injury endothelial cells directly.
