摘要
实验研究了纤维素酶和中性蛋白酶共同水解双孢蘑菇蛋白的水解工艺,以α-氨基氮含量为指标,确定了纤维素酶和中性蛋白酶的最佳水解条件为:先加纤维素酶,料液比1∶20,加酶量250U/g,初始pH值6.0,酶解温度55℃,酶解时间120min;后加中性蛋白酶,加酶量1500U/g,初始pH值6.5,酶解温度45℃,酶解时间150min。经双酶水解后,α-氨基氮含量可达36.96mg/g。
In this study the Agaricus bisporus protein is hydrolyzed by two kinds of enzymes:cellulase and neutral protease. The conditions of extraction are screened with the content of α-amino nitrogen as the marker. The optimum technical conditions are:adding cellulase first,solid to water ratio is 1 : 20, amount of cellulase 250 U/g, pH 6.0, temperature 55 ℃ to hydrolyze 120 min, then adding neutral protease,amount of meatral protease 1500 U/g, temperature 45℃,pH 6.5 to hydrolyze 150 min. After hydrolyzed by these two enzymes, the content of α-amino nitrogen is 36.96 mg/g.
